Genetic control of seed dormancy and pre-harvest sprouting in wheat Scientia Agricola
Andreoli,Claudinei; Bassoi,Manoel Carlos; Brunetta,Dionisio.
Pre-harvest sprouting (PHS) damage leads to occasional massive losses in all wheat producing areas, causing downgrading of grain quality, that severely limits end-use applications and results in substantial financial losses to farmers and food processors. Red grain color is a traditional marker for resistance to sprouting in wheat breeding programs, however red-grained genotype alone does not always guarantee effective resistance. The objective of this work was to find genes for resistance to PHS and investigate its inheritance in Brazilian wheat cultivars. Genetic variation for dormancy was investigated in the parents, F1 and 300 F2 lines derived from the cross Frontana × OR1 and its reciprocal. The germination/dormancy sprouted grains was evaluated on...

Comparative mapping reveals quantitative trait loci that affect spawning time in coho salmon (Oncorhynchus kisutch) Genet. Mol. Biol.
Araneda,Cristian; Díaz,Nelson F.; Gomez,Gilda; López,María Eugenia; Iturra,Patricia.
Spawning time in salmonids is a sex-limited quantitative trait that can be modified by selection. In rainbow trout (Oncorhynchus mykiss), various quantitative trait loci (QTL) that affect the expression of this trait have been discovered. In this study, we describe four microsatellite loci associated with two possible spawning time QTL regions in coho salmon (Oncorhynchus kisutch). The four loci were identified in females from two populations (early and late spawners) produced by divergent selection from the same base population. Three of the loci (OmyFGT34TUF, One2ASC and One19ASC) that were strongly associated with spawning time in coho salmon (p < 0.0002) were previously associated with QTL for the same trait in rainbow trout; a fourth loci (Oki10)...

One of the top priorities of the aquaculture industry is the genetic improvement of economically important traits in fish, such as those related to processing and quality. However, the accuracy of genetic evaluations has been hindered by a lack of data on such traits from a sufficiently large population of animals. The objectives of this study were thus threefold: (i) to estimate genetic parameters of growth-, yield-, and quality-related traits in rainbow trout (Oncorhynchus mykiss) using three different phenotyping technologies [invasive and non-invasive: microwave-based, digital image analysis, and magnetic resonance imaging (MRI)], (ii) to detect quantitative trait loci (QTLs) associated with these traits, and (iii) to identify candidate genes present...

Epistasis interaction of QTL effects as a genetic parameter influencing estimation of the genetic additive effect Genet. Mol. Biol.
Bocianowski,Jan.
Epistasis, an additive-by-additive interaction between quantitative trait loci, has been defined as a deviation from the sum of independent effects of individual genes. Epistasis between QTLs assayed in populations segregating for an entire genome has been found at a frequency close to that expected by chance alone. Recently, epistatic effects have been considered by many researchers as important for complex traits. In order to understand the genetic control of complex traits, it is necessary to clarify additive-by-additive interactions among genes. Herein we compare estimates of a parameter connected with the additive gene action calculated on the basis of two models: a model excluding epistasis and a model with additive-by-additive interaction effects....

The use of weighted multiple linear regression to estimate QTL-by-QTL epistatic effects Genet. Mol. Biol.
Bocianowski,Jan.
Knowledge of the nature and magnitude of gene effects, as well as their contribution to the control of metric traits, is important in formulating efficient breeding programs for the improvement of plant genetics. Information concerning a genetic parameter such as the additive-by-additive epistatic effect can be useful in traditional breeding. This report describes the results obtained by applying weighted multiple linear regression to estimate the parameter connected with an additive-by-additive epistatic interaction. Three weight variants were used: (1) standard weights based on estimated variances, (2) different weights for minimal, maximal and other lines, and (3) different weights for extreme and other lines. The approach described here combines two...

Differential allelic expression of IL13 and CSF2 genes associated with asthma Genet. Mol. Biol.
Burkhardt,Jana; Kirsten,Holger; Wolfram,Grit; Quente,Elfi; Ahnert,Peter.
An important area of genetic research is the identification of functional mechanisms in polymorphisms associated with diseases. A highly relevant functional mechanism is the influence of polymorphisms on gene expression levels (differential allelic expression, DAE). The coding single nucleotide polymorphisms (SNPs) CSF2rs25882 and IL13rs20541 have been associated with asthma. In this work, we investigated whether the mRNA expression levels of CSF2 or IL13 were correlated with these SNPs. Samples were analyzed by mass spectrometry-based quantification of gene expression. Both SNPs influenced gene expression levels (CSF2rs25882: p overall = 0.008 and pDAE samples = 0.00006; IL13rs20541: p overall = 0.059 and pDAE samples = 0.036). For CSF2, the expression...

Exploratory analysis of single nucleotide polymorphism (SNP) for quantitative traits AgEcon
Cleves, Mario A..
With the decreasing cost and the increasing ability to quickly genotype single nucleotide polymorphisms (SNP) across the human genome, large databases containing possibly hundreds of typed SNPs are becoming common in population-based studies of quantitative traits. Testing for association between individual SNPs and the quantitative trait is an important first step in the discovery of disease susceptibility SNPs. This task, however, could be time-consuming and tedious if a large number of SNPs is involved. In this article, I introduce two new commands designed to facilitate the screening and testing of multiple SNPs for possible association with quantitative traits.

Quantitative trait locus affecting birth weight on bovine chromosome 5 in a F2 Gyr x Holstein population Genet. Mol. Biol.
Gasparin,Gustavo; Miyata,Marcelo; Coutinho,Luiz Lehmann; Martinez,Mário Luiz; Silva,Marcos Vinícius G. Barbosa da; Machado,Marco Antônio; Campos,Ana Lúcia; Regitano,Luciana Correia de Almeida.
Segregation between a genetic marker and a locus influencing a quantitative trait in a well delineated population is the basis for success in mapping quantitative trait loci (QTL). To detect bovine chromosome 5 (BTA5) birth weight QTL we genotyped 294 F2 Gyr (Bos indicus) x Holstein (Bos taurus) crossbreed cattle for five microsatellite markers. A linkage map was constructed for the markers and an interval analysis for the presence of QTL was performed. The linkage map indicated differences in the order of two markers relative to the reference map (<A HREF="http://www.marc.usda.gov/">http://www.marc.usda.gov</A>). Interval analysis detected a QTL controlling birth weight (p < 0.01) at 69 centimorgans (cM) from the most centromeric marker...

Association of a locus on rat chromosome 4 with anxiety-related behaviors in two selectively bred rat lines Genet. Mol. Biol.
Hameister,Thaïs M.; Izídio,Geison S.; Valiati,Victor H.; Ramos,André.
The Floripa H and L rat lines, selected for high and low locomotion in the central aversive area of an open field, a widely used emotionality test, were proposed as a model for studying the genetic basis of anxiety. The present study aimed to verify if the QTL Ofil1, mapped to rat chromosome 4 and previously identified as being related to emotionality in another population of rats, contributes to the behavioral variability observed in the Floripa rat lines. To this purpose, rats of five generations of selective breeding were genotyped for two polymorphic markers, D4RAT59 and D4MGH27, flanking Ofil1. Changes in genotype and allele frequencies throughout generations were evaluated in both H and L lines, in order to assess if the bidirectional selection based...

Apport des informations moléculaires et cellulaires pour la caractérisation de la résistance de l'huître plate européenne vis-à-vis de la bonamiose, et pour la détection de signatures de la sélection naturelle ArchiMer
Harrang, Estelle.
The European flat oyster, an endemic species from European coasts, has been classified in the category of "endangered and/or declining species" since 2003. Indeed, the natural beds of this oyster, consumed since ancient times, have gradually been decimated by over-exploitation and by successive emergence of parasitic diseases. The parasite that causes the disease called bonamiosis has contributed to drastically reduce the French and European aquacultural production of flat oyster. Marine bivalve molluscs display two specificities that restrict possibilities to fight against diseases: they are grown in an open environment, and possess an innate immune system lacking in adaptive response. In this context, the selection of animaIs naturally resistant to...

Fine mapping and single nucleotide polymorphism effects estimation on pig chromosomes 1, 4, 7, 8, 17 and X Genet. Mol. Biol.
Hidalgo,André M.; Lopes,Paulo S.; Paixão,Débora M.; Silva,Fabyano F.; Bastiaansen,John W.M.; Paiva,Samuel R.; Faria,Danielle A.; Guimarães,Simone E.F..
Fine mapping of quantitative trait loci (QTL) from previous linkage studies was performed on pig chromosomes 1, 4, 7, 8, 17, and X which were known to harbor QTL. Traits were divided into: growth performance, carcass, internal organs, cut yields, and meat quality. Fifty families were used of a F2 population produced by crossing local Brazilian Piau boars with commercial sows. The linkage map consisted of 237 SNP and 37 microsatellite markers covering 866 centimorgans. QTL were identified by regression interval mapping using GridQTL. Individual marker effects were estimated by Bayesian LASSO regression using R. In total, 32 QTL affecting the evaluated traits were detected along the chromosomes studied. Seven of the QTL were known from previous studies using...

Quantitative trait loci mapping of pubescence density and flowering time of insect-resistant soybean (Glycine max L. Merr.) Genet. Mol. Biol.
Komatsu,Kunihiko; Okuda,Shiori; Takahashi,Masakazu; Matsunaga,Ryoichi; Nakazawa,Yoshinori.
Analysis of antibiosis resistance to common cutworm (Spodoptera litura Fabricius) in soybean (Glycine max (L.) Merr.) has progressed significantly, but the immediate cause remains unknown. We performed quantitative trait loci (QTL) analysis of pubescence density and plant development stage because these factors are assumed to be the immediate cause of resistance to cutworm. The QTLs for pubescence appeared to be identical to the previously detected the Pd1 and Ps loci controlling pubescence density. We found no candidate loci for flowering time QTLs, although one could be identical to the gene governing the long-juvenile trait or to the E6 loci controlling maturity. None of the QTLs overlapped with the QTLs for antibiosis resistance.

Increased grain dormancy in white-grained wheat by introgression of preharvest sprouting tolerance QTLs OAK
Kottearachchi, NS; Uchino, N.; Kato, Kiyoaki; Miura, Hideho; 加藤, 清明; 三浦, 秀穂.
White-grained wheat cultivars have long been recognized to be less resistant to preharvest sprouting (PHS) than the red-grained ones. Previously two QTLs for grain dormancy, QPhs.ocs-3A.1 (QPhs-3AS) and QPhs.ocs-4A.1 (QPhs-4AL) were identified in a highly dormant Japanese red wheat, Zenkoujikomugi (Zen). Aiming at improvement of PHS tolerance in white-grained wheat, the introgression effect of these two QTLs in a white-grained population consisting of 40 recombinant inbred lines (RILs) developed from a cross between Zen and white-grained Spica was examined here. Random 20 RILs with red grains were also developed from the same cross and used as a control population. The RILs were grown in the field and in the glasshouse to evaluate the grain dormancy by...
